The functions of carnivorous mammals as seed dispersers were studied during a snow-free period of 2 years in the subalpine shrubland of northern Japan. Mammal appearances, which were recorded using three automatic cameras, were dominated by Japanese marten, Martes melampus (Wagner), followed by three carnivores (ermine, red fox, and raccoon dog) and two other mammals (field mouse and Japanese hare). Fecal analysis revealed that the martens dispersed seeds of 11 species (eight woody and three herbaceous plants), which included a vine species that is an inhabitant of lower elevation sites. Fruit weight and size, and flesh weight and volume of the fleshy-fruited species foraged by the martens were significantly greater than those that were not foraged. These facts indicate that the marten plays a key role as a relatively long-distance seed disperser for major component species with large and flesh-rich fruits in the subalpine shrubland. 相似文献

Clitocypin, a new type of cysteine proteinase inhibitor from the mushroom Clitocybe nebularis, is a 34-kDa homodimer lacking disulphide bonds, reported to have unusual stability properties. Sequence similarity is limited solely to certain proteins from mushrooms. Infrared spectroscopy shows that clitocypin is a high beta-structure protein which was lost at high temperatures. The far UV circular dichroism spectrum is not that of classical beta-structure, but similar to those of a group of small beta-strand proteins, with a peak at 189nm and a trough at 202nm. An aromatic peak at 232nm and infrared bands at 1633 and 1515cm(-1) associated with the peptide backbone and the tyrosine microenvironment, respectively, were used to characterize the thermal unfolding. The reversible transition has a midpoint at 67 degrees C, with DeltaG=34kJ/mol and DeltaH=300kJ/mol, and is, unusually, independent of protein concentration. The kinetics of thermal unfolding and refolding are slow, with activation energies of 167 and 44kJ/mol, respectively. A model for folding and assembly is discussed. 相似文献

A novel electrochemical reflection cell combining electrochemical techniques and spectroscopy which uses a solid gold working electrode as an optical mirror is described. This cell can be used at path lengths as low as a few micrometers and thus is suitable for ultraviolet/visible (UV/Vis) and infrared spectroscopy even for aqueous solutions and suspensions. The cell was designed for small sample volumes of only a few microliters, thus reducing the effort for sample preparation. Due to the short path length of some micrometers, the entire volume is within the Nernst diffusion layer, hence resulting in fast equilibration. Evaluation of the technique is described with direct electrochemistry of horse heart cytochrome c at the gold electrode modified with 4,4'-dithiodipyridine. Cyclic voltammograms indicate rapid and reversible electrochemistry with the correct midpoint potential (52 mV vs Ag/AgCl/3 M KCl). Chronoamperometry and coulometry confirm rapid and complete oxidation and reduction; the cell volume can be entirely fully reduced within less than 10-20 s. Spectroscopy in the UV/Vis region, with potentials at the working electrode stepped between -390 and 390 mV, show perfect titration of the cytochrome c heme bands. A Nernst fit of the alpha band absorption, with redox potential Em and number of electrons n left as parameters, yields a midpoint potential of 49 mV and n=0.9. The potential of this cell in the investigation of biological electron transfer reactions and in the study of bioenergetic systems is discussed. 相似文献

Infrared and Raman spectroscopy were applied to identify restraints for the structure determination of the 20 amino acid loop between two beta-sheets of the N-terminal region of the PsbQ protein of the oxygen evolving complex of photosystem II from Spinacia oleracea by restraint-based homology modeling. One of the initial models has shown a stable fold of the loop in a 20 ns molecular dynamics simulation that is in accordance with spectroscopic data. Cleavage of the first 12 amino acids leads to a permanent drift in the root means square deviation of the protein backbone and induces major structural changes. 相似文献

Jaguars Panthera onca inhabiting tropical or subtropical evergreen moist forest have often been classified as opportunistic predators because they consume prey relative to its availability. However, these studies failed to address simultaneously the distribution of predator and prey through time and space, which may lead to an incomplete or erroneous understanding of jaguar foraging strategies. In this study, we reconstructed jaguar diet from scat, and used camera traps to investigate jaguar prey availability and the distribution of jaguar and its prey through space and time. We focused our examination of predator–prey temporal and spatial relations on forest infrastructure comprising man-made paths, small mammal trails, tapir Tapirus bairdi trail and trail-less, forested areas as they represent distinct habitats for prey selection. Overall, we observed high overlap between the prey used and available, suggestive of opportunistic foraging. However, jaguars exhibited selective tendencies in discriminating between larger prey. Jaguars used collared peccary Tayassu tajacu greater than its availability, while preying upon the equally abundant and similarly distributed white-lipped peccary Tayassu pecari and tapir less than predicted based upon availability. Armadillo Dasypus novemcinctus and paca Agouti paca , 56.6% of total consumption, were consumed relative to availability but exhibited low spatial overlap with jaguar. Armadillo and paca used trail-less, forested areas and small mammal trails not used by jaguar and were photographed more frequently at greater distances from man-made paths, major thoroughfares for jaguars. This study suggests that although forest jaguars use prey relative to its abundance, jaguars may rely on foraging strategies other than chance encounters for exploiting prey. 相似文献

We employ an implementation of rapid-scan Fourier transform infrared (FT-IR) microspectroscopic imaging to acquire time-resolved images for assessing the non-repetitive reorganizational dynamics of aqueous dispersions of multilamellar lipid vesicles (MLVs) derived from distearoylphosphatidylcholine (DSPC). The spatially and temporally resolved images allow direct and simultaneous determinations of various physical and chemical properties of the MLVs, including the main thermal gel to liquid crystalline phase transition, comparisons of vesicle diffusion rates in both phases and the variation in lipid bilayer packing properties between the inner and outer lamellae defining the vesicle. Specifically, in the lipid liquid crystalline phase, the inner bilayers of the MLVs are more intermolecularly ordered than the outer regions, while the intramolecular acyl chain order/disorder parameters, reflecting the overall characteristics of the fluid phase, remain uniform across the vesicle diameter. In contrast, the lipid vesicle gel phase displays no intermolecular or intramolecular dependence as a function of distance from the MLV center. 相似文献

Probing protein conformational changes plays a crucial role in protein structure and function studies. However, the lack of efficient biophysical techniques makes it difficult to obtain the distinct behaviors of different secondary structure elements in a protein upon perturbation. This paper presents a discussion of the two major problems, the effect of sidelobes and different half-width at half-height (HWHH) values, encountered in quantitative second-derivative infrared (QSD-IR) spectroscopy and introduces the development of two criteria for checking the validity of the results obtained using the QSD-IR method. It was found that neither the sidelobes nor the HWHH significantly affected the quantitative result of protein conformational changes by using poly-l-lysine and hemoglobin as model proteins. A case study of bovine serum albumin (BSA) thermal aggregation suggested that the thermal transition of BSA was a process involving sequential events, and the two helical components were found to have a distinct response to heat perturbation. These results were confirmed by two-dimensional infrared correlation spectroscopy and by results in literature, suggesting that the QSD-IR method might be a potentially powerful tool to probe the distinct response of different secondary structures to perturbation. 相似文献

This paper reviews the role of soil respiration in determining ecosystem carbon balance, and the conceptual basis for measuring and modeling soil respiration. We developed it to provide background and context for this special issue on soil respiration and to synthesize the presentations and discussions at the workshop. Soil respiration is the largest component of ecosystem respiration. Because autotrophic and heterotrophic activity belowground is controlled by substrate availability, soil respiration is strongly linked to plant metabolism, photosynthesis and litterfall. This link dominates both base rates and short-term fluctuations in soil respiration and suggests many roles for soil respiration as an indicator of ecosystem metabolism. However, the strong links between above and belowground processes complicate using soil respiration to understand changes in ecosystem carbon storage. Root and associated mycorrhizal respiration produce roughly half of soil respiration, with much of the remainder derived from decomposition of recently produced root and leaf litter. Changes in the carbon stored in the soil generally contribute little to soil respiration, but these changes, together with shifts in plant carbon allocation, determine ecosystem carbon storage belowground and its exchange with the atmosphere. Identifying the small signal from changes in large, slow carbon pools in flux dominated by decomposition of recent material and autotrophic and mycorrhizal respiration is a significant challenge. A mechanistic understanding of the belowground carbon cycle and of the response of different components to the environment will aid in identifying this signal. Our workshop identified information needs to help build that understanding: (1) the mechanisms that control the coupling of canopy and belowground processes; (2) the responses of root and heterotrophic respiration to environment; (3) plant carbon allocation patterns, particularly in different forest developmental stages, and in response to treatments (warming, CO2, nitrogen additions); and (4) coupling measurements of soil respiration with aboveground processes and changes in soil carbon. Multi-factor experiments need to be sufficiently long to allow the systems to adjust to the treatments. New technologies will be necessary to reduce uncertainty in estimates of carbon allocation, soil carbon pool sizes, and different responses of roots and microbes to environmental conditions. 相似文献

Photolytic release of ATP from inactive P(3)-[1-(2-nitrophenyl)]ethyl ester of ATP (NPE-caged ATP) provides a means to reveal molecular interactions between nucleotide and enzyme by using infrared spectroscopy. Reaction-induced infrared difference spectra of bovine intestinal alkaline phosphatase (BIAP) and of NPE-caged ATP revealed small structural alterations on the peptide backbone affecting one or two amino-acid residues. After photorelease of ATP, the substrate could be hydrolyzed sequentially by the enzyme producing three Pi, adenosine, and the photoproduct nitrosoacetophenone. It was concluded that NPE-caged ATP could bind to BIAP prior to the photolytic cleavage of ATP and that Pi could interact with BIAP after photolysis of NPE-caged ATP and hydrolysis, yielding infrared spectra with distinct structure changes of BIAP. This suggests that the molecular mechanism of ATP hydrolysis by BIAP involved small structural adjustments of the peptide backbone in the vicinity of the active site during ATP hydrolysis which continued during Pi binding. 相似文献
